Description

G-Biosciences' Amine Magnetic Beads are 1 μm homogeneous magnetic beads with amine (-NH2) surface functional groups. Magnetic beads consist of a single crystal Fe3O4 core and a dextran coating. Through chemical modification of dextran, the main amino group (-NH2) is attached to the magnetic beads through a short hydrophilic linker. The hydrophilic surface ensures the excellent dispersibility and easy handling of the magnetic beads in various buffers.

Magnetic beads with surface-active amino groups can immobilize ligands such as proteins, peptides, carbohydrates, or other target-specific molecules. Ligand immobilization can be achieved by reductive amination of aldehydes or ketones without prior activation of the bead surface. Alternatively, carbodiimide crosslinkers can bind ligands to amines through the carboxyl groups of the amines. Finally, amine-reactive heterobifunctional crosslinkers can be used to introduce additional functional groups for ligand coupling.

Carbodiimide activation of the carboxyl group produces a very unstable intermediate that undergoes rapid hydrolysis, implying that the ligand needs to be added rapidly. Alternatively, a two-step protocol using n-hydroxysuccinimide (NHS) can be used to produce unstable intermediates with longer reaction times.
