TNFRSF9 (also known as ILA; 4-1BB; CD137; CDw137) belongs to the tumor necrosis factor (TNF) receptor superfamily. TNFRSF9 is a type II membrane glycoprotein. TNFRSF9 is an important activated immune checkpoint molecule on the surface of T cells.
TNFRSF9 is ubiquitously expressed on mouse and human CD4+ and CD8+ T cells. In addition, TNFRSF9 is also expressed in activated natural killer (NK) and dendritic cells (DC) in mice. Human TNFRSF9 is also expressed in DCs, monocytes, liver cancer cells, and blood vessels of patients with malignant tumors. TNFRSF9 can be rapidly expressed on CD4+ and CD8+ T cells after antigen exposure, and activated TNFRSF9 regulates T cell-mediated immune responses by transmitting activation, proliferation or apoptosis signals between immune cells. As a newly discovered immunostimulatory molecule, TNFRSF9's potential strength should not be underestimated.
