Analysis of Alanine Aminotransferase (ALT/GPT) by Ultraviolet-visible Spectrophotometer (CAT#: STEM-MB-1349-LGZ)

Introduction

Sensitivity: 1.26 IU/L
Detection range: 1.26-72.3 IU/L
Precision: inter-lot difference of 8.199999999999999%, intra-lot difference of 4.3%
Detection equipment: Ultraviolet-visible Spectrophotometer
Detection wavelength: 505 nm

Principle

At 37℃ and pH7.4, ALT catalyzed the amino conversion reaction between alanine and α-ketoglutaric acid to produce pyruvate and glutamic acid. After the reaction time, phenylhydrazine was added to form phenylhydrazone with pyruvate. Phenylhydrazone was reddish brown in alkaline condition.

Applications

It is suitable for the detection of ALT activity in c.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: It is suitable for the detection of ALT activity in serum (plasma), animal tissues, cells and other samples
