Analysis of APBB1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2195-LGZ)

Introduction

Official Full Name: amyloid beta precursor protein binding family B member 1
Also known as: RIR; FE65; MGC:9072
The protein encoded by this gene is a member of the Fe65 protein family. It is an adapter protein localized in the nucleus. It interacts with Alzheimer's disease amyloid precursor protein (APP), transcription factors CP2/LSF/LBP1, and low-density lipoprotein receptor-associated protein. APP serves as a cytoplasmic anchor site that prevents nuclear translocation of gene products. This encoded protein may play an important role in the pathogenesis of Alzheimer's disease. It is thought to regulate transcription. Furthermore, it has also been observed to prevent cell cycle progression by downregulating the expression of thymidylate synthase. Multiple alternatively spliced transcript variants encoding different isoforms have been described for this gene.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
