Analysis of AQP7 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1943-LGZ)

Introduction

Official Full Name: aquaporin 7<br />Also known as: AQP7L; AQPap; GLYCQTL<br />This gene encodes a member of the aquaporin family of water-selective membrane channels. The encoded protein localizes to the plasma membrane and allows the movement of water, glycerol and urea across the cell membrane. This gene is highly expressed in adipose tissue, where the encoded protein promotes glycerol efflux. In the proximal straight tubule of the kidney, the encoded protein localizes to the apical membrane, preventing glycerol excretion into the urine. The encoded protein is present in spermatozoa, but also in testicular and epididymal spermatozoa, suggesting that it plays an important role in later stages of spermatogenesis. Alternative splicing of this gene results in multiple transcript variants encoding different isoforms. The gene is located near a related aquaporin gene on chromosome 9. Multiple pseudogenes of this gene have been identified.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
