Introduction

Official Full Name: Rho guanine nucleotide exchange factor 11<br />Also known as: GTRAP48; PDZ-RHOGEF<br />Rho GTPases play an important role in many cellular processes initiated by extracellular stimuli through G protein-coupled receptors. The encoded protein likely forms a complex with the G protein and stimulates rho-dependent signaling. In rats, a similar protein interacts with the glutamate transporter EAAT4 and regulates its glutamate transport activity. Expression of the rat protein induces reorganization of the actin cytoskeleton and its overexpression induces membrane ruffling and formation of filamentous feet. Two alternative transcripts encoding different isoforms have been described.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
