Analysis of Aspartate Aminotransferase (AST/GOT) Activity by Fluorescence Microplate Reader (CAT#: STEM-MB-1514-LGZ)

Introduction

Sensitivity: 0.03 U/L
Detection range: 0.03-1.5 U/L
Precision: inter-lot difference of 7.3%, intra-lot difference of 2.2%
Detection equipment: Fluorescence Microplate Reader
Excitation wavelength: 535 nm
Emission wavelength: 587 nm

Principle

Glutamic oxaloacetic acid catalyzed substrate reaction to oxaloacetic acid, oxaloacetic acid decarboxylated to pyruvate. Pyruvate is oxidized to produce hydrogen peroxide, which causes the fluorescent probe to fluoresce. The increase of fluorescence intensity in the system was proportional to AST activity.

Applications

It was used to detect the activity of AST in serum, plasma, animal tissue, cell and cell supernatant samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Determination of fluorescence intensity.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: serum, plasma, animal tissue, cell and cell supernatant samples
