Analysis of BUB1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1469-LGZ)

Introduction

Official Full Name: BUB1 mitotic checkpoint serine/threonine kinase
Also known as: BUB1A; BUB1L; hBUB1; MCPH30
This gene encodes a serine/threonine protein kinase that plays a central role in mitosis. The encoded protein functions in part to phosphorylate members of the mitotic checkpoint complex and activate the spindle checkpoint. The protein also plays a role in repressing activation of the anaphase promoting complex/ring body. The protein may also play a role in the DNA damage response. Mutations in this gene are associated with aneuploidy and several forms of cancer. Alternate splicing results in multiple transcript variants

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Carcinoma of colon

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
