Introduction

Official Full Name: CREB regulated transcription coactivator 2
Also known as: TORC2; TORC-2
This gene encodes a transducer member of the transcriptional coactivator family that regulates cAMP response element binding protein activity. These proteins promote the transcription of genes targeted by cAMP response element binding proteins and thus play an important role in many cellular processes. Under basal conditions, the encoded protein is phosphorylated by AMP-activated protein kinase or salt-inducible kinase and sequestered in the cytoplasm. Upon activation by cAMP or elevated calcium, the encoded protein translocates to the nucleus and increases the expression of target genes. A single nucleotide polymorphism in this gene may increase the risk of developing type 2 diabetes. A pseudogene of this gene is located on the long arm of chromosome 5.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
