Tight junctions are a mode of intercellular adhesion of epithelial or endothelial cell sheets that form a continuous seal around the cell and serve as a physical barrier preventing the free passage of solutes and water through the paracellular space. This immunoglobulin superfamily gene member encodes a protein that is an important regulator of tight junction assembly in epithelial cells. In addition, the encoded protein may serve as (1) a receptor for reovirus, (2) a ligand for the integrin LFA1 involved in leukocyte trafficking, and (3) a platelet receptor. Multiple 5' alternatively spliced variants encoding the same protein have been identified, but their biological validity has not been determined.
