Principle

Formate dehydrogenase (FDH) can catalyze the reaction of formic acid and NAD+ to produce NADH. Under the action of PMS, NADH transfers electrons to WST-8 to produce yellow products with characteristic absorption peaks at 450 nm. The presence of NAD(P)H in the sample will produce certain background interference, so it is necessary to set a control hole to eliminate such interference during the determination process.

Applications

For the determination of formate content in tissue samples and serum (plasma) and other liquid samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
