Principle

GDH catalyzes the dehydrogenation of glutamate and reduces NAD+ to NADH. The generated NADH reduces WST-8 to orange substance under the action of electron transport substance. This substance has the maximum absorption peak detected at about 450nm, which reflects the enzyme activity of the sample according to the rate of absorbance change.

Applications

It is suitable for detecting the activity of glutamate dehydrogenase (GDH) in serum (plasma), urine and animal tissue samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
