Introduction

Official Full Name: hemoglobin subunit delta
The delta (HBD) and beta (HBB) genes are normally expressed in adults: two alpha chains plus two beta chains make up HbA, which makes up approximately 97% of total hemoglobin in normal adult life. Two alpha chains and two delta chains make up Hb-2, which together with HbF make up the remaining 3% of adult hemoglobin. Five β-like globin genes were found in a 45 kb cluster on chromosome 11 in the following order: 5′-epsilon-gamma-Agamma-delta-beta-3′. Mutations in the -globin gene are associated with -thalassemia.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
