Analysis of Hexokinase (HK) Activity by Enzyme-labeled Instrument (CAT#: STEM-MB-1482-LGZ)

Introduction

Sensitivity: 5.93 U/L
Detection range: 5.93-40.11 U/L
Precision: inter-lot difference of 9.1%, intra-lot difference of 3.3%
Detection equipment: Enzyme-labeled Instrument
Detection wavelength: 440-450 nm

Principle

Hexokinase converts glucose to glucose 6-phosphate, which is catalyzed by an enzyme, while converting NAD+ to NADH, which reduces WST-8 to an orange-yellow substance with an electron-coupled reagent, with a maximum absorption peak at around 450 nm. The color of orange substance produced in the reaction system can reflect the level of hexokinase activity in the sample.

Applications

It is suitable for detecting hexokinase (HK) enzyme activity in animal tissue and cell samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: animal tissue and cell samples
