Introduction

Official Full Name: potassium voltage-gated channel subfamily A member 10<br />Also known as: Kcn1; Kv1.8<br />Potassium ion channels are the most complex class of voltage-gated ion channels from both functional and structural point of view. Their diverse functions include regulation of neurotransmitter release, heart rate, insulin secretion, neuronal excitability, epithelial electrolyte transport, smooth muscle contraction, and cell volume. Four sequence-related potassium channel genes—shaker, shaw, shab, and shall—were found in Drosophila and each had human orthologs. This gene encodes a member of the potassium channel, voltage-gated, shaker-related subfamily. This member contains six transmembrane domains with a shaker-type repeat in the fourth segment. It is specifically regulated by cGMP and is thought to mediate the action of substances that increase intracellular cGMP. This gene has no introns and is clustered on chromosome 1 with the KCNA2 and KCNA3 genes.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
