Analysis of Klotho (Human) by MSD Hypersensitive Multi-Factor Electrochemiluminescence Analyzer (CAT#: STEM-MB-0380-LGZ)

Introduction

Klotho is a transmembrane protein that regulates the sensitivity of organisms to insulin. It has β-glucuronidase activity and can hydrolyze β-glucuronic acid, which is related to human aging. Circulating klotox in plasma decreases gradually with age. β-Klotsole will cooperate with FGF receptors, and can bind to FGF19, FGF20, FGF23 and other fibroblast growth factors (FGF). The mechanism of action of Kloxol is still unknown, but it is known that it can regulate the calcium balance of cells by increasing the expression of TRPV5 and reducing the expression of TRPC6. In addition, Kloxol also increases the expression of inward rectifier ion channel ROMK.

Principle

MSD electrochemiluminescence technology is mainly based on ultra-sensitive multi-factor electrochemiluminescence analyzer MES0 Sector600 or Quickplex SQ120. Through dot matrix technology, the detection of 10 indicators per well can be realized in the 96-hole graphite electrode plate. It can also be customized to realize the screening detection of 100 index wells in a 24-well plate.

Applications

Kidney Injury, Metabolic, Toxicology

Procedure

1. Coat the capture antibody on the well plate.
2. Add samples/calibrators.
3. Add detection antibody.
4. Read the plate and analyze the data.

Materials

• Sample Type: Plasma, Serum
