Analysis of KRT19 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2485-LGZ)

Introduction

Official Full Name: keratin 19<br />Also known as: K19; CK19; K1CS<br />The protein encoded by this gene is a member of the keratin family. Keratin is an intermediate filament protein responsible for the structural integrity of epithelial cells and is subdivided into cytokeratin and hair keratin. Type I cytokeratins consist of acidic proteins arranged in pairs of heterokeratin chains. Unlike related family members, this smallest known acidic cytokeratin does not pair with basal cytokeratins in epithelial cells. It is specifically expressed in the periderm, the transient superficial layer that wraps the developing epidermis. Type I cytokeratins accumulate in a region of chromosome 17q12-q21.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
