Analysis of MRPL20 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1358-LGZ)

Introduction

Official Full Name: mitochondrial ribosomal protein L20
Also known as: L20mt; MRP-L20
Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and assist in protein synthesis within mitochondria. Mitochondrial ribosomes consist of a small 28S subunit and a large 39S subunit. They are estimated to be composed of 75% protein and rRNA compared to prokaryotic ribosomes, whose proportions are reversed. Another difference between mammalian ribosomes and prokaryotic ribosomes is that the latter contain 5S rRNA. The proteins that make up the mitochondrial body vary widely in sequence and sometimes in biochemical properties across species, making sequence homology difficult to identify. This gene encodes a 39S subunit protein. A pseudogene corresponding to this gene was found on chromosome 21q. Alternative splicing results in multiple transcript variants encoding different isoforms.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
