Analysis of MYH11 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2473-LGZ)

Introduction

Official Full Name: myosin heavy chain 11
Also known as: AAT4; FAA4; SMHC; SMMHC; VSCM2; SMMS-1
The protein encoded by this gene is smooth muscle myosin, which belongs to the myosin heavy chain family. The gene product is a subunit of a hexameric protein consisting of two heavy chain subunits and two pairs of non-identical light chain subunits. It is a major contractile protein that converts chemical energy into mechanical energy through the hydrolysis of ATP. Chromosomal rearrangements involving this gene have been associated with acute myeloid leukemia of the M4Eo subtype. Mutations in this gene are associated with visceral myopathy, megacystis-microcolon-intestinal hypoperistalsis syndrome 2, and familial thoracic aortic aneurysm 4.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
