Analysis of NUF2 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0508-LGZ)

Introduction

Official Full Name: NUF2 component of NDC80 kinetochore complex
Also known as: CDCA1; CT106; NUF2R
The gene encodes a protein that is highly similar to yeast Nuf2, a component of a conserved protein complex associated with centromeres. Yeast Nuf2 disappears from the centromere when it loses its connection to the spindle pole body during meiotic prophase and plays a regulatory role in chromosome segregation The encoded protein was found to be associated with the centromeres of mitotic HeLa cells, suggesting that this protein is a functional homologue of yeast Nuf2. Alternative splice transcript variants encoding the same protein have been described.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
