Analysis of PDGFRA Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1836-LGZ)

Introduction

Official Full Name: platelet derived growth factor receptor alpha
Also known as: CD140A; PDGFR2; PDGFR-2
This gene encodes a cell surface tyrosine kinase receptor for a member of the platelet-derived growth factor family. These growth factors are mitogens for mesenchymal cells. The identity of the growth factor bound to the receptor monomer determines whether the functional receptor is a homodimer or a heterodimer, consisting of platelet-derived growth factor receptor alpha and beta polypeptides. Studies have shown that the gene plays a role in organ development, wound healing and tumor progression. Mutations in this gene are associated with idiopathic hypereosinophilic syndrome, somatic and familial gastrointestinal stromal tumors, and a variety of other cancers.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gastrointestinal stromal tumor

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
