The gene, a member of the period gene family, is expressed in a circadian pattern in the suprachiasmatic nucleus, the major circadian pacemaker of the mammalian brain. Genes in this family encode circadian components of locomotor activity, metabolism, and behavior. The gene is upregulated by the CLOCK/ARNTL heterodimer, but then interacts with CLOCK/ARNTL through the PER/CRY heterodimer, inhibiting this upregulation in a feedback loop. Polymorphisms in this gene may increase the risk of certain cancers and have been linked to sleep disturbances.
