Analysis of RAMP1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1645-LGZ)

Introduction

Official Full Name: receptor activity modifying protein 1<br />The protein encoded by this gene is a member of the RAMP family of single-transmembrane domain proteins, known as receptor (calcitonin) activity-modifying proteins (RAMPs). RAMPs are type I transmembrane proteins with an extracellular N-terminus and a cytoplasmic C-terminus. RAMPs are required for the transport of calcitonin receptor-like receptors (CRLRs) to the plasma membrane. CRLR is a receptor with seven transmembrane domains that acts as both a calcitonin gene-related peptide (CGRP) receptor and an adrenomedullin receptor, depending on which members of the RAMP family are expressed. In the presence of this (RAMP1) protein, CRLR functions as a CGRP receptor. The RAMP1 protein is involved in the terminal glycosylation, maturation and presentation of CGRP receptors to the cell surface. Alternative splicing results in multiple transcript variants encoding different isoforms.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
