Analysis of RBMXL2 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2109-LGZ)

Introduction

Official Full Name: RBMX like 2
Also known as: HNRPGT; HNRNPGT; HNRNPG-T
This gene belongs to the HNRPG subfamily of ubiquitously expressed heteronuclear ribonucleoproteins (hnRNPs). hnRNPs are RNA-binding proteins that complex with heterogeneous nuclear RNA (hnRNA). These proteins associate with pre-mRNAs in the nucleus and appear to affect pre-mRNA processing and other aspects of mRNA metabolism and trafficking. Although all hnRNPs are present in the nucleus, some appear to shuttle between the nucleus and cytoplasm. hnRNP proteins have unique nucleic acid binding properties. The protein encoded by this gene has two RRM domains that bind RNA. This gene is intronless and is thought to be derived from a processed retrocon. However, unlike many retro-derived genes, this gene is not a pseudogene. The encoded protein has similarity to HNRPG and RBMY proteins, and may replace the function of HNRPG protein in meiotic prophase or act as a germ cell-specific splicing regulator. It is mainly localized in the nucleus of meiotic spermatocytes. This gene is a candidate gene for autosomal male infertility.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
