Analysis of SHC1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0662-LGZ)

Introduction

Official Full Name: SHC adaptor protein 1<br />Also known as: SHC; SHCA<br />The gene encodes three major isoforms that differ in activity and subcellular location. Although all three proteins are adapter proteins in the signal transduction pathway, the longest (p66Shc) may be involved in the regulation of lifespan and the role of reactive oxygen species. The other two isoforms, p52Shc and p46Shc, link activated tyrosine receptor kinases to the Ras pathway by recruiting the GRB2/SOS complex. p66Shc is not involved in the activation of Ras. Unlike the other two isoforms, p46Shc targets the mitochondrial matrix. Several transcript variants of this gene have been found encoding different isoforms.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
