Analysis of TNFRSF1B Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1257-LGZ)

Introduction

Official Full Name: TNF receptor superfamily member 1B
Also known as: p75; TBPII; TNFBR; TNFR2; CD120b; TNFR1B; TNFR80; TNF-R75; p75TNFR; TNF-R-II
The protein encoded by this gene is a member of the TNF receptor superfamily. This protein forms a heterocomplex with TNF receptor 1, mediating the recruitment of two anti-apoptotic proteins c-IAP1 and c-IAP2 with E3 ubiquitin ligase activity. The function of IAPs in TNF receptor signaling is unclear, however, c-IAP1 is thought to enhance TNF-induced apoptosis by mediating the ubiquitination and degradation of TNF receptor-associated factor 2, an anti-apoptotic signal. Knockout studies in mice also showed that the protein protects neurons from apoptosis by stimulating antioxidant pathways.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
