Analysis of TP73 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1442-LGZ)

Introduction

Official Full Name: tumor protein p73<br />Also known as: P73; CILD47<br />This gene encodes a member of the p53 family of transcription factors involved in cellular responses to stress and development. It maps to a region on chromosome 1p36 that is frequently deleted in neuroblastoma and other tumors and is thought to contain multiple tumor suppressor genes. Evidence that this gene is monoallelic expressed (likely from the maternal allele) supports its notion that it is a candidate gene for neuroblastoma. Many transcript variants of this gene have been identified due to alternative splicing and/or use of alternative promoters, but the biological validity and full-length nature of some variants have not been determined.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
