Analysis of TSHB Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1168-LGZ)

Introduction

Official Full Name: thyroid stimulating hormone subunit beta
Also known as: TSH-B; TSH-BETA
The Four Human GlyCoprotein Hormones Chorionic Gonadotropin (CG), Luteinizing Hormone (LH), FolLICLE Stimulating Hormone (FSH), and Thyroid Stimulating Hormone (TS H) Are Dimers consisting of Alpha and Beta Subunits that are associated noncovalently. The alpha subunits of these hormones are identical, however, their beta chains are unique and confer biological specificity. Thyroid-stimulating hormone has the function of controlling the structure and metabolism of the thyroid gland. The protein encoded by this gene is the beta subunit of thyrotropin. Mutations in this gene are associated with congenital central and secondary hypothyroidism and Hashimoto's thyroiditis. Alternative splicing of this gene results in multiple transcript variants.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
