Analysis of WNT10A Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1513-LGZ)

Introduction

Official Full Name: Wnt family member 10A<br />Also known as: OODD; SSPS; ECTD16; STHAG4<br />The WNT gene family consists of structurally related genes encoding secretory signaling proteins. These proteins have been implicated in tumorigenesis and several developmental processes, including the regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It is strongly expressed in promyelocytic leukemia and Burkitt lymphoma cell lines. Furthermore, it and another family member, WNT6 gene, were strongly co-expressed in colorectal cancer cell lines. The overexpression of this gene may play a key role in carcinogenesis by activating the Wnt-β-catenin-TCF signaling pathway. This gene clusters with the WNT6 gene in the chromosome 2q35 region.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
