Bicinchoninic Acid (BCA) Assay

The bicinchoninic acid (BCA) assay is a biochemical assay for determining the total concentration of protein in a solution (0.5 μg/mL to 1.5 mg/mL). The total protein concentration is exhibited by a color change of the sample solution from green to purple in proportion to protein concentration, which can then be measured using colorimetric techniques.

Procedure
  • Reagents
    Serially dilute bovine serum albumin (BSA) standards.
    Prepare BCA working solution.
  • Assay
    Add each dilution of protein standard and test protein sample to a microplate or test tube.
    Add BCA working solution separately.
    After sealing, incubate at 37°C for 30-60 minutes.
  • Analysis
    Measure the absorbance of the sample at 562 nm.
    Calculate the protein concentration of the sample based on the standard curve and the sample volume used.
Features
  • This method is simple and fast, four times faster than the lowry method.
  • The sensitivity is high, the minimum detection amount is 0.5µg.
  • It has good stability and good linearity in the range of 20µg/ml~2000µg/ml.
  • It is economical and practical, and the volume of the sample to be tested can be 1µl~20µl.
Application
  • Widely used for protein concentration determination.
Precautions
  • When using BCA protein determination method, the color will be deepened with the extension of time, and the speed of color reaction is related to the temperature, so it is necessary to maintain the timing and constant temperature to ensure accurate quantification.
  • Low temperature or long-term storage, if it is found that BCA reagent A or B precipitation occurs. Incubate at 37ºC and stir to dissolve fully. If bacterial contamination is found, it should be discarded to avoid affecting the experimental results.
  • The experimenter should operate the specification to improve the accuracy of sample loading.
  • The corresponding standard curve should be made for each measurement, because the color reaction is related to the change of temperature and time, and the standard curve should be made every time for accurate protein quantification.
