Highlights

The main components of Western and IP cell lysate are 20mM Tris (pH7.5), 150mM NaCl, 1% Triton X100, and various inhibitors such as sodium pyrophosphate, βglycerophosphate, EDTA, Na3VO4, leupeptin, etc. It can effectively inhibit protein degradation and maintain the original protein-protein interaction.
The protein samples obtained by Western and IP cell lysis buffer can be used to measure the protein concentration with BCA protein concentration assay kit.
Due to the high concentration of interfering substances such as Triton X100, the Bradford method cannot be used to determine the protein concentration of the samples lysed by this lysate.
