The main components of NP40 lysate are 1% NP40, 50 mM TrisHCl (pH 7.4), 150 mM NaCl, and Sodium Orthovanadate, Sodium Fluoride, EDTA, etc. The mild cleavage method is beneficial to maintain the original protein structure and maintain the original protein-protein interaction.
This product needs to be used together with conventional protease inhibitors to achieve better extraction results; this product contains phosphatase inhibitors, which can be used to extract phosphorylated proteins.
The protein sample obtained by lysing with NP40 lysis buffer can be used to measure the protein concentration with the BCA protein concentration assay kit. Due to the high concentration of interfering substances such as detergents, the Bradford method cannot be used to determine the protein concentration of the sample lysed by this lysing solution.
