Description

The carboxy-coupling resin consists of 6% cross-linked agarose and covalently linked diaminodipropylamine (DADPA) to generate free primary amines at the ends of the long spacer arms. This resin is also known as EAH-Agarose.

Molecules, including proteins, peptides, and nucleic acids, are covalently coupled to free primary amines, and stable columns are ideal for affinity purification of antibodies and other interaction partners.

Molecules can be coupled to free amines via a variety of amine reaction methods; however, the use of carbodiimide EDCs allows coupling of free carboxyl groups. The resulting amide bond is highly stable, greatly reducing the chance of leaching of the affinity tag. Long spacer arms reduce steric hindrance, ensuring greater binding of protein and antibody during affinity purification.

The carboxyl-coupled resin has a very high capacity for immobilizing DADPA and provides 60 μmol amine loading per milliliter of resin. This high capacity provides unparalleled efficiency and capability in immobilizing carboxyl-containing proteins.
