Column Chromatography Technology

Column Chromatography Technology is a distribution method with distribution balance as the mechanism. The chromatographic system consists of two phases, one is the stationary phase and the other is the mobile phase. When the two phases move relative to each other, the differences in the distribution equilibrium properties of the components contained in the mixture are used repeatedly, and finally, the purpose of separation is achieved. This technique can be used on a small scale as well as a large scale to purify materials that can be used in future experiments. It is a common method for the purification and separation of organic or inorganic substances.

Procedure
  • Adsorbent filling
    The adsorbent is filled into the column as the stationary phase. The stationary phase is wetted with the help of solvent. The mobile phase or eluent is either a solvent or a mixture of solvents.
  • Sample introduction
    The mixture of compounds to be separated is added from the top of the column without disturbing the top level.
  • Sample elution
    The eluent is added from the top of the column, adjusting the appropriate flow rate. When the compound to be separated arrives at the end of the column, it is collected in a clean test tube.
