By cutting the entire genomic DNA of a cell with restriction endonuclease, a large number of genomic DNA fragments can be obtained, which are then ligated to a vector and transformed into a bacterium, allowing the host bacterium to grow into a clone. In this way, each cell within a clone has a specific genomic DNA fragment contained on the vector, and the entire clonal population contains the total sum of all the genomic fragments of the genome called the genomic library.
