Genomic Library Construction to store all gene of specific organism (CAT#: STEM-MB-0031-WXH)

Introduction

By cutting the entire genomic DNA of a cell with restriction endonuclease, a large number of genomic DNA fragments can be obtained, which are then ligated to a vector and transformed into a bacterium, allowing the host bacterium to grow into a clone. In this way, each cell within a clone has a specific genomic DNA fragment contained on the vector, and the entire clonal population contains the total sum of all the genomic fragments of the genome called the genomic library. Gene library = genomic library + cDNA library .

Applications

• Storage and amplification of all or part of the genome of a specific organism
• Retrieve any DNA fragment or target gene from a genomic library when necessary

Procedure

1.Preparation of vector DNA:
Enzymatic reaction of vector DNA - Purification of vector arm - Vector dephosphorylation treatment
2.Ligation and packaging
3.Infection of host bacteria
4.Preservation and amplification of genomic library
5.Titer measurement
6.Preservation

Notes

Customer provides: blood or cells from animals, fresh plant leaves, live insects or purified DNA samples.
