Hydroxyapatite [Ca10(PO4)6(OH)2] is an inorganic pure-phase chromatography resin composed of spheres calcined at high temperatures. Its crystal structure matches that of bone hydroxyapatite, featuring a stable spherical structure and uniform particle sizes. Hydroxyapatite interacts with biomolecules through multiple modes, enabling two simultaneous separation mechanisms: cation exchange and calcium metal affinity. The cation exchange is driven by the negatively charged phosphate groups on hydroxyapatite. Additionally, carboxyl and phosphorus clusters on biomolecules form strong bonds with calcium on hydroxyapatite through metal affinity.
Due to these unique separation mechanisms, hydroxyapatite is essential in today's demanding downstream processes and plays a crucial role in biopharmaceuticals, including antibodies, vaccines (bacterial polysaccharides, pneumonia polysaccharides, and viruses), nucleic acids, enzymes, and recombinant proteins.
