Description

The immobilized imine biotin resin consists of imine biotin, a cyclic guanine analog of biotin, covalently coupled to 6% cross-linked agarose. This resin allows the purification of avidin, streptavidin and neutravidin with their subsequent gentle elution using non-denaturing elution buffer.

The normal biotin-avidin complex requires a strong denaturing buffer, 8M guanidine·HCl, to denature avidin and release biotin, which obviously destroys the native and functional aspects of avidin. The iminobiotin-avidin complex is formed at pH 9.5 and can be separated at pH 4.0 with a mild elution buffer consisting of 50 mM ammonium acetate, pH 4.0, 0.5M NaCl.
