Description

MagINDEX™ rProtein A/G Magnetic Sepharose is an affinity chromatography medium designed for fast, simple, one-step purification of a broad range of immunoglobulins, as well as fusion proteins containing Fc regions.

Proprietary MagIndex™ technology facilitates encapsulation of superparamagnetic cores in a mechanically stable agarose matrix, which is optimal absorbance for high-yield protein purification with minimal background.

Superparamagnetic cores have a very fast magnetic response in a magnetic field, but have no memory when the magnetic force is removed. The agarose-based encapsulation provides a strong shield that protects the core well in a wide range of reagents commonly used in protein purification. In addition, as an ideal protein purification matrix, agarose matrix can achieve high-purity and high-capacity purification of antibodies and Ig fragments in high-throughput screening and various preparation scales.

Protein A is a bacterial cell wall protein isolated from Staphylococcus aureus that binds to the Fc region of most immunoglobulins through interaction with the heavy chain. Native protein A contains five high-affinity IgG-binding domains and many domains with unknown functions. Native protein G has three IgG binding domains as well as albumin and cell surface binding sites, while recombinant protein G has only an Ig recognition domain and no other non-specific sequences.

Recombinant protein A/G is a recombinant chimeric protein consisting of the IgG binding domains of protein A and protein G. Protein A/G contains 5 binding domains of protein A and 2 binding domains of protein G. Protein A/G binding is less pH-dependent than protein A, but otherwise has the additivity of protein A and protein G. Therefore, Protein A/G is an ideal protein to bind the broadest IgG subclass in rabbit, mouse, human, and other mammalian samples.
