Partition Chromatography Technology

Partition Chromatography Technology is a kind of chromatographic method which separates different components by the difference in their partition properties. A polar solvent immobilized on support is called a stationary phase, and it does not move. Elution with another, less polar solvent, called the mobile phase, is always mobile. Both phases are in liquid form. The components of a mixture are separated if their partition coefficient between the solvents is sufficiently different.

The most commonly used carriers include silica gel, diatomaceous earth (Kresel guhr, celite), and cellulose, among others. Silica gel must be deactivated by impregnation with water or other polar solvents before use. The stationary liquid phase and the mobile liquid phase must be immiscible with each other. Otherwise, the two phases merge into one and no partition can occur. The general rule is that the more soluble the solute particles are in the mobile phase, the smaller the partition coefficient and the faster the elution from the column. From this, it can be seen that the more solute particles dissolved in the stationary phase, the larger the partition coefficient, the slower the downward movement spee.

Types

These are some important types of partition chromatography technology.

Column Chromatography Technology – In this technique, liquid stationary phase is taken on silica gel or alumina cellulose powder.

Thin Layer Chromatography Technology – In this technique, a very thin layer of stationary phase is taken.

Paper Chromatography Technology – In this technique, liquid stationary phase is taken on chromatography paper made up of cellulose which is a polar substance. Water molecules get trapped in it.
