Description

The ProteIndexTM Streptavidin Agarose 6FF provides a simple, fast and reliable method for the purification of biotin and biotinylated substances. Streptavidin ligands are recombinant versions of native proteins derived from Streptomyces bacterium that have a high affinity for biotin. Immobilized streptavidin is on a highly cross-linked 6% agarose matrix support, so it can be used for affinity chromatography to purify biotin and biotinylated substances. Due to the strong interaction between streptavidin and biotin, the elution of biotin and biotinylated substances usually requires harsh denaturing conditions. Streptavidin also binds to 2-iminobiotin and iminobiotinylated substances in a pH-dependent manner. When the pH was greater than 9.0, the affinity interaction between streptavidin and imine biotin was stronger, and the affinity interaction decreased significantly with the decrease of pH. Thus, imine-biotinylated species can be captured on immobilized streptavidin at pH 9.5 - 11.0 and eluted at pH 4.0.

Additionally, immobilized streptavidin on a matrix support can be used to purify biomolecules capable of interacting with and forming complexes with biomolecules for bioanalysis. For example, antigens can be purified by immunoreactive complexes, whose specific biotinylated antibodies are captured onto streptavidin Sepharose 6FF resin, and the antigens can be eluted when they are separated from the complexes.

The base matrix of Streptavidin Sepharose 6FF is highly cross-linked 6% agarose with excellent flow properties and high mechanical stability. This resin is well suited for batch/gravity cleanup, centrifugation procedures, or FPLC processes on automated systems.
