BMP-2 belongs to the transforming growth factor-beta (TGF-β) superfamily of structurally related signaling proteins. BMP-2 is a potent osteoinductive cytokine that, in conjunction with collagen and synthetic hydroxyapatite as bone-conductive carriers, induces the formation of bone and cartilage. In addition to its osteogenic activity, BMP-2 plays a crucial role in heart morphogenesis and is expressed in various tissues such as the lungs, liver, spleen, prostate, ovaries, and small intestine.
The functional form of BMP-2 is a 26 kDa protein composed of two identical 114 amino acid peptide chains (monomers) connected by a single disulfide bond. Each BMP-2 monomer is expressed as the C-terminal part of a precursor polypeptide, which also includes a 23 amino acid signal sequence for secretion and a 259 amino acid propeptide. After dimerization of this precursor, the covalent bond between the propeptide (also a dimer) and the mature BMP-2 ligand is cleaved by furin-type proteinases.
The lyophilized form can be stored at 2-8°C for 24 months. For aliquoting, it can be reconstituted using sterile water, saline, culture medium, or PBS. After reconstitution, it is recommended to divide it into smaller portions and store at -20°C for 6 months or -80°C for 12 months, with a final diluted concentration of no less than 100 μg/mL, while avoiding repeated freeze-thaw cycles.
