Description

The Recombinant Human Holo-Transferrin Protein manufactured by Seafrom is suitable for studying iron transport and cellular processes.
Transferrin is a blood plasma glycoprotein that binds iron, regulating the levels of free iron in biological fluids. This glycoprotein is believed to have originated from an ancient gene duplication event, resulting in the generation of homologous C and N-terminal domains, each capable of binding one ferric iron ion. The primary function of Transferrin is to transport iron from the intestine, reticuloendothelial system, and liver parenchymal cells to all proliferating cells in the body. It is a complex protein composed of alpha-helices and beta sheets, forming two domains—the first located in the N-terminus and the second in the C-terminus. These N- and C-terminal domains are represented by globular lobes, with an iron-binding site situated between them.
The liver is the primary source of transferrin production, although other sources, such as the brain, also produce this glycoprotein. Transferrin is associated with the innate immune system, found in the mucosa, where it binds iron, creating an environment with low free iron that hinders bacterial survival in a process known as iron withholding. The level of transferrin decreases during inflammation. Transferrin transports iron ions from the hemolymph into eggs during the vitellogenic stage. Transferrins are iron-binding transport proteins that can bind two Fe(3+) ions in association with the binding of an anion, usually bicarbonate. Their primary role is to transport iron from sites of absorption and heme degradation to locations for storage and utilization. Serum transferrin may also have an additional role in stimulating cell proliferation. The pH is reduced by hydrogen iron pumps, causing transferrin to release its iron ions. The receptor is then transported through the endocytic cycle back to the cell surface, ready for another round of iron uptake. Each transferrin molecule can carry two iron ions in the ferric form.
The lyophilized formulation can be stored at 4°C for 24 months, and the reconstituted liquid can be stored at -20°C for 6-12 months, avoiding repeated freeze-thaw cycles.
