Description

The Recombinant Human IL-18 Protein (E.coli) manufactured by Seafrom is suitable for pro-inflammatory effects and a role in immune responses.
Interleukin-18 (IL-18), also known as gamma interferon-inducing factor, is an inflammatory cytokine belonging to the interleukin-1 family. It can be produced by macrophages and other cell types. This cytokine induces the production of interferon-gamma by T cells. When IL-18 combines with IL-12, it can suppress IL-4-dependent IgE and IgG1 production while promoting the production of IgG2a by B cells. IL-18 binding protein (IL-18BP) specifically interacts with this cytokine, providing negative regulation of its biological activity. IL-18 is also an IL-1-like cytokine that requires cleavage by the enzyme caspase-1 to become active. This activity can increase the expression of CD4-positive cells, IgE in the presence of IL-4 and STAT6. In in vitro experiments, IL-18 and T cell receptor-mediated stimulation can induce the further development of natural CD4-positive T cells into IL-4-producing cells.
Hence, caspase-1 and IL-18 play important regulatory roles in IgE production in vivo and provide a potential therapeutic target for allergic disorders. IL-18 produced in early synovial tissue cultures and synovial fibrous tissues can be alternatively regulated by TNF-α and IL-1β, indicating that the expression of mononuclear factors in the synovium can promote the development of Th1 cells. Additionally, the synergistic effects of multiple factors such as IL-18, IL-12, and IL-15 are crucial for maintaining Th1 cell responses and mononuclear factor production in rheumatoid arthritis (RA).
The lyophilized formulation can be stored at 2-8°C for 24 months. For reconstitution, it can be dissolved in injection-grade water, physiological saline, culture medium, or PBS. After dissolution, it should be divided into smaller aliquots and stored at -20°C for 6 months or at -80°C for 12 months, with a final concentration after dilution not lower than 100 μg/mL. Avoid repeated freeze-thaw cycles.
