Single-chain Antibody (scFv) Production (CAT#: STEM-MB-0135-WXH)

Introduction

Single-chain antibody (scFv) is a small molecule composed of the variable region of the antibody heavy chain and the variable region of the light chain connected by a peptide chain, and is the smallest functional structural unit with antibody activity . Single-chain antibodies can be expressed by expression systems, and can also be prepared by phage display technology.
scFv has the advantages of small molecular weight, strong penetrating power, and high specificity, and has important applications in targeted therapy, imaging diagnosis, intracellular immunity, and biological detection.

Applications

• Targeted therapy: Drugs and toxins can be linked to scFv to specifically kill target cells.
• Imaging diagnosis: Due to the strong penetrating power of scFv, the distribution index in tumor tissue is higher than that of intact antibody molecules.
• Radiation Imaging: Radioactive nuclides are discharged quickly, with little harm to the body, and can be used for tumor imaging and localization diagnosis; Intracellular immunity: single-chain antibodies against viral.
• Proteins can be cloned and expressed in cells, which can be used for viral infectivity disease diagnosis and treatment.

Procedure

1. Preparation of scFv gene library
2. Transfer scFv gene into phage
3. Infecting host bacteria
4. Obtain scFv antibody library
5. Screen the phage antibody library to obtain high-affinity scFv
