Description

Biotin is a 244Da vitamin (vitamin H) molecule with extraordinary binding affinity for avidin (Ka=10¹â µM-¹) and streptavidin (Ka=10¹â µM-¹). Biotin and avidin interact rapidly. Once the bond is established, it can survive 3M Guanidine-HCl and extreme pH levels. The biotin-avidin bond can only be reversed by denaturing the avidin protein molecule with 8M guanidine-HCl at pH 1.5, or by boiling in SDS Page Sample Loading Buffer.

Streptavidin is a tetrameric protein containing four biotin-binding sites. Streptavidin is similar in many respects to avidin, but it contains no carbohydrates and has a slightly lower molecular weight (approximately 60 kDa). Streptavidin (isoelectric pH 5) is much less soluble than avidin in aqueous buffers; however, streptavidin binds biotin very similarly to avidin.

The advantage of streptavidin is that the absence of carbohydrates significantly reduces the amount of nonspecific binding.

The streptavidin used for immobilization on porous 6% cross-linked agarose was in recombinant form with a mass of 53 kDa and near neutral pH. Streptavidin is covalently coupled to agarose, resulting in minimal leaching, and is stable from pH 2 to 11.

Streptavidin resins are designed for single-step, small and large-scale affinity purification of proteins, antibodies and other molecules with biotin tags. The resin is also useful for assay development and immunoprecipitation of proteins using biotinylated antibodies.
