Introduction

.Chelate, any of a class of coordination or complex compounds consisting of a central metal atom attached to a large molecule, called a ligand, in a cyclic or ring structure. Chelates are more stable than nonchelated compounds of comparable composition, and the more extensive the chelation—that is, the larger the number of ring closures to a metal atom—the more stable the compound. In medical practice, chelating agents, particularly salts of EDTA, or edetic (ethylenediaminetetraacetic) acid, are widely used for direct treatment of metal poisoning because they bind the toxic metal ions more strongly than do the vulnerable components of the living organism. Chelating agents are also employed as extractants in industrial and laboratory separation of metals and as metal-ion buffers and indicators in analytical chemistry. Many commercial dyes and a number of biological substances, including chlorophyll and hemoglobin, are chelate compounds. 2C12 is a monoclonal antibody that can be prepared by BALB/c mouse injection of a Pb (II) -chelate complex covalently coupled to a carrier protein. 2C12 can recognize the Pb (II) -cyclohexyldiethylenetriamine pentaacetic acid complex




Principle

KinExA is a two-stage analysis system. In the first stage, a number of solutions are prepared, where one partner remains constant (constant binding partner, or CBP) and the other (titrant) is variable, usually serially diluted. As the titrant is added, the free CBP decreases and is analysed by a sophisticated and precise microfluorescence measurement device. The signal generated can be mathematically related to the affinity (KD) of the two molecules for each other, as well as the kinetic parameters of binding (kon) and dissociation (koff).

Procedure

1. preparation of the functionalized beads which will capture the analyte for measuremen.
2. preparation of a series of solutions consisting of a constant initial concentration of one component of the binary reaction and serial dilutions of the other reactant. The component that is kept constant is the constant binding partner (CBP) , and is the one which will be analyzed.
3. each reaction mixture is sampled and the fluorescence of free CBP bound to the capture beads is obtained for subsequent numerical analysis.
