Analysis of AKT3 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0795-LGZ)

Introduction

Official Full Name: AKT serine/threonine kinase 3
Also known as: MPPH; PKBG; MPPH2; PRKBG; STK-2; PKB-GAMMA; RAC-gamma; RAC-PK-gamma
The protein encoded by this gene is a member of AKT, also known as PKB, the serine/threonine protein kinase family. AKT kinase is thought to be a regulator of cellular signaling by insulin and growth factors. They are involved in a variety of biological processes, including cell proliferation, differentiation, apoptosis, tumorigenesis, and glycogen synthesis and glucose uptake. This kinase has been shown to be stimulated by platelet-derived growth factor (PDGF), insulin, and insulin-like growth factor 1 (IGF1). In addition, splice transcript variants encoding different isoforms have been described.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
