Analysis of EIF2AK2 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1803-LGZ)

Introduction

Official Full Name: eukaryotic translation initiation factor 2 alpha kinase 2
Also known as: PKR; PRKR; DYT33; LEUDEN; EIF2AK1; PPP1R83
The protein encoded by this gene is a serine/threonine protein kinase that is activated by autophosphorylation upon binding to dsRNA. The activated form of the encoded protein phosphorylates the translation initiation factor EIF2S1, thereby inhibiting protein synthesis. This protein is also activated by manganese ions and heparin. The encoded protein plays an important role in the innate immune response against a variety of DNA and RNA viruses.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
