Analysis of RGS1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0596-LGZ)

Introduction

Official Full Name: regulator of G protein signaling 1
Also known as: 1R20; BL34; IER1; IR20; HEL-S-87
This gene encodes a member of the G-protein signaling regulator family. The protein is located on the cytoplasmic side of the plasma membrane and contains a conserved 120 amino acid motif called the RGS domain. This protein attenuates the signaling activity of G proteins by binding to activated GTP-bound G α subunits and acting as a GTPase activating protein (GAP), increasing the conversion rate of GTP to GDP. This hydrolysis allows the Gα subunit to associate with the Gβ/γ subunit heterodimer to form an inactive G protein heterotrimer, thereby terminating the signal.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
