Analysis of RHOG Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-2108-LGZ)

Introduction

Official Full Name: ras homolog family member G
Also known as: ARHG
This gene encodes a member of the Rho family of small GTPases that cycles between inactive and active GTP-bound states and acts as a molecular switch in signal transduction cascades. Rho proteins promote reorganization of the actin cytoskeleton, regulating cell shape, attachment and motility. The encoded protein facilitates the translocation of functional guanine nucleotide exchange factor (GEF) complexes from the cytoplasm to the plasma membrane, where it activates the Ras-associated C3 botulinum toxin substrate 1, promoting Phytophthora formation and cell migration. Two associated pseudogenes were identified on chromosomes 20 and X.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
